Walk in there like you own the place. In order to draw attention (the more people that see it the better the result) without it looking like you're trying to make a scene, ask in a slightly raised voice "which one of you is [insert faggots name here]?". March straight up to his desk and be like "why have you been telling people that you slept with my wife?".
The object of the excercise is to make him squirm as much as possible, so you're going to ask him lots of questions, push him in to trying to justify his actions. Things like "who have you told", "why did you think it was ok to do it", "did you think I was just going to let you spread rumours about my family", that kind of stuff. The endgame is making him apologise - to you, to your wife, to the people he's been lying to.
Be sure to remain calm, don't get angry, don't raise your voice, don't make threats. Work to a plan, and have a clear idea of how you're going to end the confrontation - you're disrupting the workflow in someone else's business, so you need to make sure you've done everything you want to before you get asked to leave/slung out.
Not only will you totally destroy the guys reputation, he's going to get chewed out by HR over it, and people from that company will still be talking about it in 2020.
